Biologische Wirkwertbestimmung von Ginkgo-Extrakten

Abstract: Prof. Dr. Dr. h. c. H. Wagner Institut für Pharmazeutische Biologie der Universität München Zusammenfassung Die Messung der PAF-induzierten Thrombozytenaggregation wird als Invitro-Methode zur Bestimmung der PAF-antagonistischen Wirkung der Ginkgolide vorgestellt. Anhand der IC50 Werte werden Wirkwerte als äquieffektive Ginkgolid-B-Gehalte verschiedener Ginkgo-Extrakte ermittelt. Bei Kenntnis der Ginkgolid-Anteile am Ginkgolid-Gemisch und des Wirkungsverhältnisses der Hauptginkgolide böte sich eine Umrechnung des äquieffektiven Ginkgolid-B-Gehalts auf den Gesamtginkgolid-Gehalt an. Wenngleich durch die biologische Wirkwertbestimmung eine chemische Gehaltsbestimmung nicht ersetzt zu werden braucht so ist sie jedoch - sofern die klinische Relevanz der PAF-antagonistischen Wirkung der Ginkgolide für Ginkgo-Extrakte gesichert ist - als therapierelevante Standardisierung von Bedeutung. Auf der Basis klinischer Erfahrungen sollte in Zukunft eine Mindestforderung für den Wirkwert erarbeitet werden der den heutigen Qualitätsanforderungen an Ginkgo-Präparate entspricht. Determination of the Biological Effective Value of Ginkgo Extracts: The present paper reports n measurements of PAF-induced thrombocyte aggregation for in vitro determination of the PAF-antagonistic effect of the ginkgolides. The effective values of various ginkgo extracts can be correlated with equieffective ginkgolide-B contents on the basis of IC50 readings. Where the ginkgolide fractions in the ginkgolide mixture and the ratio of effect of the principal ginkgolides are known it is possible to express the equieffective ginkgolide-B content as a fraction of the total ginkgolide content. Although a chemical assay does not need to be replaced by a determination of the biological effective value it is still important for standardization for treatment purposes - as far as the clinical relevance of the PAF-antagonistic effect of the ginkgolides in respect of ginkgo extracts has been proven. A minimum requirement for the effective value that corresponds to the current demands on the quality of ginkgo preparations must be laid down an the basis of clinical experience. ab
