Lokale Tumortherapie mit dem Differenzierungsinduktor Tributyrin: Grundlagen der Therapie und Vorstellung einer Phase-I-Studie

Abstract: In a great number of studies butyric acid (a non-toxic fatty acid that arises as a by-product of microbial fermentation) has been shown to induce differentiation and to inhibit proliferation of various malignant cells. This paper reviews the published experimental and clinical applications of butyric acid in malignant disease. We present the main features of a clinical phase-1 trial studying local application of tributyrin in patients with non-resected rectal carcinoma (inoperable primary tumor or local recurrence). The study is mainly aimed at determining the maximum tolerated dose by means of dose escalation. In addition, as far as possible within the design of this study, antineoplastic effects of the treatment are evaluated.
