Influence of the duration of pretreatment with very low concentrations of cadmium on cytotoxicity induced by the metal and on cultured kidney tubular cells

Abstract: Cadmium is one of the most frequent encountered polluting agents in the environment. Recent automatic methods have been developed to quantitate cytotoxicity of cells, cultured in microtiter plates, induced by xenobiotics and in particular heavy metals. Since cadmium is a pollutant with kidney tropism, its effect was studied in vitro on a kidney cell-line (LLCPK1). The aim of the present study was to evaluate the effect of pre- treating cultured LLCPK1 cells for 2-5 days with dilutions of cadmium ranging from 10-14 to 10-20 M on the cytotoxicity caused by a 24-h exposure to the same agent at 2-5x10-5 M. Cell viability was measured by the neutral red ant MTT tests. Cultures pre-incub s suggest that there are cellular mechanisms able to induce the synthesis of protective substances as metallothioneins.
