Histamine liberation in vitro and comparison of the effect in vivo of dilutions of histamine and apis to that of cimetidine and ketotifene

Abstract: In a previous report, we showed that the 4CH dilution of histamine or the 7CH dilution of Apis have a protective effect in the polymyxin (PMB) - ulcer model in rats. Using this same model, we have studied the effect of various substances on histaminemia after a single injection i.p. of 2mg/kg PMB to confirm the results obtained. Male Wistar rats weighing 180+/- 10g were dcvided into 5 lots of 10 . Thirty minutes before treatment,thcy received: either a single in jection of a 4CH dilution of histamine,60mg/kg of cimctidine or 1mg/kg ketotifen or 2 injections daily for 2x3 days of,a 4CH dilution of histamine. Histamine was measured by automatic spectrofluorimetric method 30 min after egranulation at doses which in fact caused histamine release. In light of these results, one of the hypothesis of the mechanism of action mentioned previously does not seem to be supported.
