, , , ,Abstract: In this randomized, double-blind, placebo-controlled, parallel group clinical trial 81 male and female patients with chronic venous insufficiency (stages I and II) participated. Aim of the trial was to test the clinical efficacy of a buckwheat herb/troxerutin combination by measuring the reduction in lower leg edema. After a 2-week run-in phase 37 patients received 3x2 tablets containing 500 mg buckwheat herb and 30 mg troxerutin daily; 44 patients received placebo tablets. Thee active treatment period was 12 weeks and was followed by a 4-week post observation period. The main outcome measure was the lower leg volume of the more seriously affected leg determined by ultrasound. The treatment group demonstrated a mean decrease in lower leg volume of 62 ml (from 2051 ± 427 to 1989 ± 377 ml). In contrast, the placebo group did not show any changes (from 2086 ± 485 to 2083 ± 508 ml). The difference between the treatments was statistically significant (p < 0.05). Subjective clinical symptoms assessed by a symptom score were also significantly improved in the treatment group as compared with placebo. The diameter of the femoral vein showed only slight changes. The treatment with a combination of buckwheat herb and troxerutin was well tolerated and significantly reduced edema development as well as clinical symptoms in patients with chronic venous insufficiency.
