Zur Standardisierung vom Mistelpräparaten

Abstract: Remedies of mistletoe are used in the therapy of cancerous diseases. They contain substances of high biological activity, i.e. Iectins, viscotoxins, oligosaccharides, and lipids. Not only one of these substances alone, but acting in union of all components of a given mistletoe preparation is responsible for the effectiveness against cancer. Therefore standardization only based on lectins isn't suffficient for protection pharmaceutical quality which can be achieved only by employing process-standardization. Only by standardization the complete manufacturing process including harvesting times, recipe, manufacturing steps and validated controll-methods preparation of standardized reinedies of mistletoe with reliable action in tnan is possible, and the content of active compounds (e.g. Iectins) will be constant from batch to batch.Two mistletoe extracts prepared from leaves and shoots, respectively, and one commercial available preparation were analysed in a collaborative study by 6 laboratories. Different usually employed methods of determination lectins, i.e. hemagglutination-, cytotoxicity-, ELISAand ELLA-test, were used. Hemagglutination- and cytotoxicity-test react as nonspecific tests not only on lectins. Therefore here the highest amounts were observed. The results of the cytotoxicitytest were more than tenfold higher than the results of the ELLA-galactose-BSA. Both ELLA-asialofetuin- and ELISA-test came to nearly the same results. The relative standard-deviation of the ELLA-asfmethod used ranged between 15 and 20%. Only ELLA-galactose-BSA is specific for mistletoe-lectin- I, not for the other lectins. The hitherto existing results of this running study showed impressively that methods of different reaction mechanisms lead to different results. The amounts of the results obtained are bound 1. to the method used, 2. to the lectinstandard-preparation used, and 3. to the preparation tested.
