Comparative studies on internalization of gold labelled mistletoe lectin I (MLI), its subunits, and of an immunotoxin into mouse L 1210 V leukemia cells

Abstract: The binding and internalization of gold-labelled mistletoe lectin I (MLI), its A (ML I-A) and B (ML I-B) subunits, as well as of an immunotoxinconsisting of an monoclonal anti L 1210 antibody and the cytotoxic Achains of ML I, were studied on murine L 1210 leukemia cells by apreembedding electron microscopic technique. We found thatreceptor-mediated endocytosis differs remarkably between the wholelectin, its subunits, and the immunotoxin. Whereas ML I, its A chain, aswell as the immunotoxin are internalized by coated pits/coated vesiclesor in combination with uncoated membranes, the B chain is exclusivelyendocytosed via uncoated deep invaginations of the cell membrane. Theendocytosis via clathrin-coated or uncoated vesicles is discussed takinginto account the binding and internalization kinetics of ligand-receptorcomplexes related with the movement by the cytoskeleton.
