TFX-Thymomodulin- Biochemische Charakterisierung und immunbiologisches Wirkprofil in vitro |

Abstract: The calf thymus extract TFX-Thymomodulin has been characterised on its biochemical properties and immunobiological effects. In different celltests (heterologous E-rosette-forming-assay, cell-proliferation-test, phagocytosis-stimulating-test) TFX was tested and in vitro-effects on T-cells (MOLT-3) and macrophages (P388D1) will be described. The HPLC-analysis uncovered the heterogeneous character of TFX, of both major components TFX* and TFXß. TFX* contains a mixture of polypeptides and peptides of various lower molecular weights. TFXß contains three peptides, which obtained the denotations ßa, ßb and ßc. By means of SDS-PAGE of ßa,b,c, ßa, ßb and ßc mainly one protein band was recovered and the molecular weight was estimated to be in the range of 4500-5000 Da and the isoelectric points of the three components ßa, ßb, and ßc, were estimated in the range of 5,2; 5,9 and 6,4. Forming heterologous E-rosettes with SRBC the biological activity of cells stimulated with the combination of the two peptides ßb and ßc was always increased. TFX ßa stimulated the proliferation-rate of T-lymphocytes (MOLT-3) and increased the phagocytosis activity of neoplasmic macrophages (P388D1) in a significant percentage. Immunological effects could not be recognized after incubation of the cells with the heterogeneous fraction TFX*.
