Pretreatment with low dose of cadmium (Cd) protects rat renal mesangial cells against the direct toxic effect of cadmium

Abstract: We have compared the response of renal mesangial cells to different doses of Cadmium. The effect of Cadmium was assessed by measuring the growth rate of an homogeneous rat renal cell line. Cell growth in the presencc of 10-6 M Cd exhibited a 37+/- 6% mortality rate within 24h. When the cells were first placed in the presence of 10-15 M or 10-20 M CdCl2 for 24,48,72,96 h and then placed in the presence of 10-6 M CdCl2, the toxic effect initially observed with this concentration was significantly reduced. This decreased in Cd cytotoxicity was also dependent on the pretreatment time. The rnaximum resistance effect was obtained after 72 h of pretreatrnent; a 45 +/-6% increase in surviv is induced by pretreatnient with Cadmium itself at doses as low as 10-15 and 10-20 M. The induction of metallothioneill is likely to be involved.
