Abstract: The usefulness of lectin affinity chromatography for the preparation ofglycoproteins is impaired by ligand release. Ligand leakage frommistletoe lectin (MLI) Sepharose 4B column was detected by 24 h skinreaction in mice and by immunoblotting. Immunoaffinity chromatographywas found to be an efficient method for the separation of lectin tracesfrom the glycoprotein fraction. A sugar concentration dependent increaseof lectin release from MLI-Sepharose 4B column was detected by a solidphase enzyme immunoassay. Author.
