SUMMARY There has been a marked and rapid increase in our understanding of the chemistry and metabolism of the polysaccharides of connective tissue. This review has described some of these advances and something of the impact of this new knowledge upon our concepts of several connective tissue diseases. Further pursuit of information in these basic areas may yield answers to many clinical problems in rheumatology. ___MH
