Donor-dependent and dose-dependent variations in the induction of T lymphocyte locomotion in a three-dimensional collagen matrix system by a mistletoe preparation (Iscador)

Abstract: Controlled activation of non-specific and specific immune defencemechanisms can beneficially manipulate the host's ability to attackmalignant cells. In this context, migration and tissue distribution ofimmunocompetent cells may be prerequisites for an efficient immunesurveillance. The effect of various non-cytotoxic concentrations of theViscum* album L. (mistletoe) preparation Iscador QuFrF on the locomotoryactivity of immunomagnetically isolated human CD4+ and CD8+ Tlymphocytes from healthy donors was investigated. Cellular migration wasexamined within a three-dimensional collagen matrix. Donor-dependentvariations in baseline activities of spontaneously locomoting T cellswere accompanied by individual response patterns of T cells fromdifferent donors in the presence of various concentrations of mistletoepreparation (0.25-2.5 mug/ml). Using the three-dimensional collagenmatrix assay an induction of locomotory activity was detected in ahighly reproducible fashion although the optimal concentration ofmistletoe preparation and the time point of maximal response wereindividual for each donor. Our data suggest that the direct stimulationof T-cell migration by mistletoe components may modulate the system ofimmune surveillance and recognition in patients under mistletoe therapy.COPYRIGHT BY Elsevier Science BV, Amsterdam, Netherlands END OF DOCUMENT
