Positive inotropic effect of MIRDTON® Experimental investigation with isolated human cardiac muscle providing a dose effect curve for MIROTONO® and digoxin

Abstract: MIROTON® enhances the isometric contractility of electrically, stimulated papillary muscle strips of moderately and highly insufficient human hearts, as well as of right atrial trabeculae of patients without heart failure. In vitro, the positive inotropic effect of MIROTON® in human papillary muscle preparations is comparable to the inotropic effect of digoxin both in terms of potency and effectiveness. The effectiveness of MIROTON® and that of digoxin were comparable to the positive inotropic effect of Ca2+ (15 mmoles/l/).MIROTON® and digoxin solutions displaced specifically bound 3H-Quabain from its receptor binding sites in a concentration-dependent manner. Hence, it appears probable that the inotropic effect achieved by MIROTON® in vitro results from an interaction with membrane-bound Na+, K+-A TPase.
