THE USE OF A DISCONTINUOUS BUFFER SYSTEM IN PREPARATIVE GEL ELECTROPHORESIS

In preparative gel electrophoresis in columns one often uses an elution buffer of the same type as the column buffer but at a higher concentration. The purpose of the increased concentration of the elution buffer is to reduce the field strength in the elution chamber and thus to prevent separated components of the sample from migrating out of the elution chamber. This method of preventing a loss of sample is however disadvantageous in that a concentration gradient of buffer ions will occur in the lower part of the gel column causing a retardation of the separated zones especially those having a lower mobility. The purpose of this paper is to describe a method to calculate a discontinuous buffer system which does not have the disadvantage mentioned above. The calculation method is based an the theory of Ornstein (1964) which in turn is based an Kohlrauschs regulating function (Kohlrausch 1897). . . . .
